One
of the method to count viable cell is namely as MPN method. MPN is an efficient
method; it could be implemented by multiple-tube lactose
fermentation technique. In this method, there is
three-stage procedure. The positive result is based on the gas
produce inside the Durham Tube and the colour changes. MPN could be implemented
by using 3 tubes or 5 tubes in one series. MPN result will help to estimate
number of cell in the sample by using standard MPN table. The three stages -- the dilution stage, presumptive stage and
confirmation stage are briefly summarized below. (For the analysis to be
accurate, three tubes are required.)
